چکیده

Previous studies have suggested that the lnotropic effects of glucagon on the guinea pig heart, but not on the rat heart, could be dissociated from its effects on cyclic AMP formation. We compared the effects of glucagon on working rat and guinea pig hearts to reinvestigate this proposed dissociation. When administered to spontaneously beating preparations, glucagon produced •imilar increases hi rate, but different increases in contractility, of rat and guinea pig hearts. ITie inotropic effects of glucagon on the rat heart -were greater in magnitude than those on the guinea pig heart. Glucagon (10"* to 3 x 10"' M) increased left ventricular pressure (LVP) and the rate of pressure development (dP/dt), and reduced the time-to-peak pressure (TPP) of both rat and guinea pig spontaneously beating hearts. Studies in which the chronotropic responses to glucagon (10"' M) were duplicated electronically showed that the inotropic effects on the spontaneously beating guinea pig heart were entirely frequency-dependent, whereas those on the rat heart were not. In addition, glucagon (10" M) markedly increased LVP, dP/dt, and ventricular cyclic AMP levels of the paced rat heart, but had no effect on any of these variables in the paced guinea pig heart. The results do not support the proposed dissociation between cyclic AMP generation and positive inotropic responses of the guinea pig heart to glucagon. They suggest further that the guinea pig heart exhibits regional differences in sensitivity to glucagon. Ore Rea 49: 21&-23S, 1981

Dissociations between Changes in Myocardial Cyclic Adenosine Monophosphate

The relationship between changes in the myocardial concentration of adenosine 3':5'-cyclic monophosphate (cyclic AMP) and cardiac contractility was studied in guinea pig and rat myocardium. When isolated perfused guinea pig hearts were perfused with 10"M papaverine, a potent inhibitor of cyclic AMP phosphodiesterase activity, myocardial cyclic AMP concentration increased significantly from 1.7 ...

The Role of Nitric Oxide and Prostaglandins in the Effect of Adenosine on Contractility, Heart Rate and Coronary Blood Flow in Guinea Pig Isolated Heart

It is a well-established fact that adenosine and its receptor subtypes (A 1 and A ) are involved in changes of contractility, heart rate and coronary blood flow (CBF) under different circumstances. This study was conducted to evaluate the role of nitric oxide and prostaglandins in development of these changes. For this purpose, Nitro-L-Arginine methyl ester (L-NAME), and indomethacin as inhibit...

Interaction between cyclic adenosine monophosphate and cyclic gunaosine monophosphate in guinea pig ventricular myocardium.

The purpose of this study was to examine the mechanisms underlying adrenergic-cholinergic antagonism in ventricular myocardium. Myocardial contractility, cyclic adenosine monophosphate (AMP) levels, and cyclic guanosine monophosphate (GMP) levels were measure in isolated guinea pig ventricles after treatment with various inotropic agents given alone and simultaneously with acetylcholine. Acetyl...

Cyclic Adenosme Monophosphate Modulation of Slow Calcium Influx Channels in Guinea Pig Hearts

The relationship between cellular levels of cyclic adenosine monophosphate (AMP) and slow inward calcium ion (Ca) current was studied in isolated perfused guinea pig hearts in which fast sodium ion (Na) channels had been inactivated by depolarization with potassium ions (K) or blockade with tetrodotoxin. Perfusion with 22 mM K depolarized cardiac tissue to approximately -40 mv and rendered hear...
